  • Albumin Nanopartiele drug

    Principle Physical encapsulation: drugs are physically entrapped or adsorbed inside self‑assembled albumin nanoparticles.
    Binding Mode Mainly non‑covalent interactions (e.g., hydrophobic interactions, electrostatic forces, hydrogen bonding).
    Structural Form Forms nanoparticles (typically 50–200 nm) with the drug encapsulated inside.
    Drug Loading Capacity Relatively high; a single nanoparticle can encapsulate a large number of drug molecules.
    Targeting Mechanism Passive targeting (EPR effect) + active targeting (binding to receptors such as gp60 and SPARC).
    Drug Release Slow release via nanoparticle degradation and diffusion.

  • Albumin–Drug Conjugate

    Principle Chemical conjugation: drug molecules are directly “welded” onto the albumin molecule via covalent bonds.
    Binding Mode Mainly covalent bonds (e.g., using the free thiol at Cys‑34, amino groups of lysine, etc.).
    Structural Form Forms molecular conjugates; the drug is directly attached to a single albumin molecule without forming a particle.
    Drug Loading Capacity Relatively low; one albumin molecule can usually carry only a limited number of drug molecules.
    Targeting Mechanism Primarily relies on albumin’s intrinsic targeting properties (e.g., binding to SPARC).
    Drug Release Release triggered by cleavage of chemical bonds (e.g., pH‑sensitive or enzyme‑sensitive linkers).
    Preparation Process In vitro synthesis or in situ formation – either pre‑synthesised via chemical reactions, or injected as a prodrug that spontaneously binds to endogenous albumin in the bloodstream.
